中文摘要: 流感病毒的中和抗体和具有抗体依赖细胞介导的毒性杀伤能力的抗体(ADCC抗体)在抑制病毒感染宿主细胞,减缓疾病进展和清除病毒过程中发挥了关键作用。 H7N9流感患者血浆抗体的中和滴度不高,难以给机体提供保护,H7N9患者血浆抗体的ADCC特性尚不清楚。本项目以我院收集的37例H7N9流感患者发病后不同阶段的系列血浆为研究材料,采用流式细胞技术检测其介导的对HA7阳性细胞的杀伤效果,利用HA7片段酵母表面展示结合流式细胞仪筛选技术鉴定抗体的ADCC表位,并研究H7N9血浆抗体的ADCC能力及其随疾病进展的变化在病情控制和临床转归过程中的作用。本项目首次从ADCC的角度对H7N9流感患者的体液免疫机制进行研究,并首次提出对H7N9血清抗体的ADCC表位进行鉴定和验证,有望阐明H7N9流感患者的体液免疫保护机制,并可能为新型H7N9流感疫苗设计提供新的切入点。

英文摘要: Neutralizing antibody and ADCC (antibody dependent cell mediated cytotoxicity) antibody against Influenza virus not only play pivotal role in the inhibition of host cell infection, but also slow disease progression and mediate virus eradication. H7N9 patient plasma antibodies have low neutralizing titration which is not sufficient for protection. The ADCC effect of the H7N9 patient plasma antibodies is not clear. This project will use plasma samples from 37 H7N9 patients collected in our hospital as materials, we will detect the antibody mediate cytotoxic effect of HA7 positive cell by flow cytometry, we will identify the ADCC antibody specific epitopes by flow cytometry assisted cell sorting against a HA7 fragment yeast surface display library, we also study relationship between the plasma antibody’s changes of ADCC effect following disease progression and disease control and recovery. This project focuses on ADCC in the study of H7N9 patient humoral immune mechanism and raise to identify ADCC epitopes of the plasma antibodies for the first time. The completion of this study may illuminate the protective mechanism of humoral immunity and provide new concept for vaccine design of the new H7N9 influenza virus.
