中文摘要: 目前分子育种是苜蓿育种研究的热点,对直根型苜蓿的研究已在抗非生物胁迫、抗生物胁迫和品质改良等方面取得了重要进展,对根蘖型苜蓿已在形态学、遗传学、生理机制及分子生物学等方面进行了深入研究,但对根茎型苜蓿根茎性状发生的分子机制及调控根茎性状发生的相关基因仍不清楚。根茎型苜蓿作为一相对独立的紫花苜蓿种质资源,与世界上已登记苜蓿品种的遗传距离较大,是苜蓿育种和品种改良的优异种质资源。为此,本研究拟以根茎型清水紫花苜蓿为供试材料,以蒺藜苜蓿基因组为对照,采用RNA-Seq技术对根茎与非根茎样本转录组进行解析,得到与根系相关的差异表达基因;用GA对根茎型苜蓿进行诱导处理,结合cDNA-SRAP技术对与根茎性状相关的差异基因进行分离,旨在揭示控制根茎性状发生的分子机制并挖掘优异基因,不仅可促进豆类植物遗传和基因组学研究,而且对于挖掘我国根茎型苜蓿优异基因资源,促进苜蓿新品种选育和品质改良也具有重要意义。

英文摘要: At present, molecular breeding has become a hot spot in alfalfa breeding. Tap-rooted alfalfa has been achieved in abiotic stresses, biotic stress and quality improvement, and creeping rooted alfalfa has been achieved in morphology, genetic and molecular biology,molecular mechanism and regulation gene of rhizomatous rooted alfalfa is still unclear. Rhizomatous rooted alfalfa is a relative independence germplasm resource, there is bigger genetic distance between rhizomatous rooted alfalfa and international registry other' alfalfa varieties, it is an important genepools of alfalfa breeding and improved varieties. Therefore, taking the rhizomatous rooted Medicago sativa L. cv. Qingshui as the material, to take the Medicago truncatula as the control, the transcriptomic of library rhizomatous rooted and nonrhizomatous rooted will be construct, respectivelly.To analyze the library transcriptome with next generation sequences (RNA-seq). Researching the expression of differential gene through cDNA-SRAP technology in order to search the molecular mechanism of rhizomatous rooted alfalfa and finding the relevant excellent gene, which not only would promote leguminous plant genetic and genomics but also would exploit gene resources of rhizomatous rooted alfalfa, which will important to promote breeding and quality improvement of alfalfa.
