中文摘要: 猪博卡病毒(PBoV)是2009年首次在患有断奶仔猪多系统衰竭综合症的猪淋巴结中检测到的一种单股线状DNA病毒,前人研究揭示其可能是近年来仔猪腹泻病重新爆发的一个重要原因。由于缺少相关毒株,目前其致病性关键因子和机理解析等方面未见相关报道,致使PBoV处在无苗可防,无药可治状态。项目申请者前期研究分离得到一株PBoV,并证实其导致仔猪发生腹泻。进一步,本项目拟将通过人工感染仔猪,采用转录组测序技术分析仔猪感染PBoV/zj11毒株后转录水平上的差异、结合病理学分析和细胞因子变化等相关研究,确定PBoV感染致病的候选关键因子;然后构建超表达和RNAi干涉载体,通过转染猪肠上皮细胞(IPEC-J2)对其候选关键因子进行验证,从而确定PBoV致病的关键因子。通过本项目研究,可以为PBoV致病机理解析奠定基础,为后期疫苗研发和综合防控由PBoV引发的疾病提供科学依据。

英文摘要: Porcine bocavirus (PBoV), a small, single-molecule, linear DNA nonenveloped virus, was first isolated from the lymph nodes of pigs with Postweaning multisystemic wasting syndrome in 2009. Previrous studies revealed that it might be the most important reason for the renewed outbreak of pig diarrhea in recent years. Due to the lack of PBoV virulent strain, the pathogenic determinants and the pathogenic mechanism are still unclear, which implys that there hasn't any effective preventive and therapeutic strategies for PBoV. Luckly, we isolated a PBoV (PBoV/zj11 strain) and confrimed it can cause piglet diarrhea. In order to isolate the possible pathogenic determinants of PBoV/zj11 strain, we are going to use RNA-Seq technique to detect the transcriptome change in PBoV/zj11 strain-infected piglets, along with the analysis of the pathological morphology and cytokine changes.When obtained the candidated determinant genes, we will build overexpression and RNA interference vectors and transfect them to cell model (IPEC-J2) to identify their functions. Taken together, the identification of PBoV pathogenic determinants will help to elucidate the underlying pathogenic mechanism and to develop vaccine and prevention mehod for PBoV, which in the end plays an important role in the sustainable development of swine industry.
