中文摘要: 对我国杨树造成严重危害的光肩星天牛与黄斑星天牛曾经被认为是两个不同的种。除其原分布区不同,两者在形态特征、生物生态学特性等方面都非常相似,因而,它们的种间关系一直饱受争议。近年来,通过国内外学者的深入研究,证实了两者是同一种的两种型。两型光肩星天牛产生分化的主要原因很可能是历史上的地理隔离对基因交流的限制,以及对不同环境的适应性变异。目前,在两型重叠分布区发现的"中间型"现象暗示着渐渗杂交的发生。本项目借助线粒体基因序列、核基因组的SSR、AFLP等分子标记,结合群体遗传学的相关研究方法,揭示光肩星天牛的系统进化历程及基因渗透在该过程中的作用;利用cDNA-AFLP技术和比较基因组学的方法,明确与两型适应性分化相关的功能基因。最终阐明光肩星天牛的种内微进化过程和两型分化的遗传机制,为后续探讨其适应性变异的多基因调控机制的研究提供基础,同时为其它昆虫表型变异的微进化过程研究提供借鉴。

英文摘要: Anoplophora glabripennis and A. nobilis were considered as two of the most danger sibling species of poplar in China. They display quite similar morphologial, physiological and behavioural characteristics. Recently, A. nobilis was placed in synonymy with A. Glabripennis. This synoymy has supported by crossing-mating studies, morphological analyses and molecular comparisons. Geographic isolation and heterogeneous environment were resulted in divergence of the two strains. Although primaeval allopatric, in some regions the ranges of two members of A. Glabripennis meet or overlap now. Morphologically intermediate individuls were found in overlap regions, which implying the possibility of intraspecific introgression. In this project, several molecular markers (mitochondrial DNA sequences, mtDNA; microsatellite, SSR; Amplified fragment length polymorphism, AFLP) will be used to illustrate the genetic variation of A. glabripennis. The evolution dynamic and the influence of introgressive in shaping genetic structure pattern of this insect will be assessed. To identify the suite of genes involved in the differentiation of A. glabripennis, a genome-wide cDNA-AFLP gene expression assay will be conducted as an platform to screen for candidate genes that are differentially expressed between the two strains.
