中文摘要: 本项目围绕水稻生长素调控细胞分裂素葡萄糖基转移酶基因COG1表达,决定水稻根系建成的信号调控机制进行深入和全面的探索和阐述。通过分离鉴定可能的调控COG1分生区特异表达的水稻ARF突变体,明确调控COG1分生区特异表达的ARF转录因子以及相关的调控路径;全面剖析生长素诱导COG1表达,调节细胞分裂素活性转换在水稻侧生根和不定根分生组织形成和维持中的功能及其在调节后续次生器官形成中的机制。利用已获得的COG1-RNAi和过表达转基因株系,系统研究生长素调控COG1表达水平,精细调节水稻根系发育关键调控因子WOX11, ARL1, CRL5等表达的规律和机制,建立水稻侧生根和不定根分生组织形成和维持以及根系发育的调控路径。本项将从生长素调控水稻内源细胞分裂素糖基化修饰这一全新的角度,揭示生长素调节细胞分裂素细胞内平衡和信号转导效率,决定根部分生组织形成和维持的分子机制。

英文摘要: The objective of this project is to dissect the regulatory mechanisms of auxin in modulating the expression of Cytokinin-O-Glucosyltransferase1 (COG1) during development and maintenance of root meristems in rice. By identifying ARF(s), we are going to define the regulatory pathway of auxin signaling in regulation of the meristematic expression of COG1. The effects of up- or down-regulation of COG1 expression will be examined on the expression patterns of WOX11, ARL1, CRL5, the important genes involving the initiation and development of lateral roots and crown roots, respectively, by examining the rice transgenic plants of COG1-RNAi and over-expression. Our project is going to reveal a new cross-talk mechanism in which auxin regulates the root meristem formation through modulating the activation of COG1, an auxin-responsive gene critical for cytokinin homeostasis and signal transduction in rice.
