中文摘要: 污染环境微生物原位修复具有高效、安全、成本低、无二次污染等优点,是有发展前景的污染修复方式。然而,目前对微生物原位修复机理的研究还不够透彻,对于微生物原位修复过程中菌株生物学行为的研究还比较缺乏。六六六是上世纪七十年代前世界范围内曾广泛使用的一种有机氯杀虫剂,由于其高毒性和长残留性,造成了严重的环境污染,尤其一些高浓度污染场地给当地带来了严重危害,问题亟待解决。本项目拟以菌株Sphingobium japonicum UT26为研究对象,借助454高通量测序及实时荧光定量PCR等技术,结合一些传统的研究方法,深入研究菌株在六六六污染场地微生物修复过程中的存活和定殖、降解基因的表达以及对土壤微生物区系的影响等生物学行为,并将其与修复效果相偶联,阐明六六六污染场地微生物修复效果背后的微生物学和分子生物学原理,为该菌株在六六六污染场地修复中的应用提供理论参考,具有较好的科学价值和应用价值。

英文摘要: Microbial remediation is a prospects way to remove pollutions from contaminated environments, because of its efficient, safe and low-cost, no secondary pollution, etc. However, the study of Microbial remediation mechanisms is not enough now. It especially lack of the study on biological behavior during the remediation procedure. HCH is an organochlorine insecticide which was widely used in worldwide before the seventies of the last century. Because of its high toxicity and long residual, it caused serious environmental pollution, especially in some contaminated sites where with high concentration of HCH, and need to be solved urgently. This project intends to use the strain Sphingobium japonicum UT26 as the object to study the Microbial remediation of the HCH contaminated sites with 454 high-throughput sequencing and real-time fluorescence quantitative PCR technology, as well as some traditional research methods. In this project, we will deeply study the biological behavior such as the survival of strain UT26 and the degrading genes expression level and the impact on soil microbial Community in the HCH contaminated sites to clarify the principle of microbiology and molecular biology behind the BHC contaminated sites microbial remediation effects, which will provide theoretical reference for the use of strain UT2
