中文摘要: 成功检测到大鼠骨髓间充质干细胞(BMSCs)表达神经肽受体,BMSCs成骨分化中神经肽受体表达增加。神经肽通过影响细胞周期相关调控基因蛋白的表达,促进对BMSCs的增殖作用。神经肽通过作用于VCAM-1诱导BMSCs发生迁移作用。神经肽作用于BMSCs,能促进成骨细胞特征性物质AP及BGP合成,能明显提高骨形态发生蛋白2(BMP2),血管内皮生长因子(VEGF)的蛋白表达,其中降钙素基因相关肽(CGRP)能促进VEGF,CGRP受体,focal adhesion kinase(FAK),VEGF receptors 1,VEGF receptors 2在HUVECs中的表达,从而显著促进血管内皮细胞发生增殖,迁移,体外成管作用,CGRP体外成血管作用甚至强于低浓度VEGF。血管束,感觉神经束植入组织工程骨能明显促进骨修复及提高组织工程骨神经肽和神经肽受体的表达。血管束组织工程骨各时间点中VEGF的表达量高于对照组。因此神经肽可同时作用于骨髓基质干细胞和血管内皮细胞上的神经肽受体,从增加成骨活性,促进血管新生等多方面对组织工程骨新生起到了积极的促进作用。

英文摘要: Neuropeptide receptors have been detected in rat BMSCs,the expression of neuropeptide receptors increased in the osteoblast-induced groups.Neuropeptide had direct effects on the proliferation of BMSCs,the regulation of cell cycle proteins is one of the mechanisms.Neuropeptide had a significantly enhanced role in promoting cell migration resulting from the increase expression of VCAM-1.Neuropeptide can also stimulate the expression of AP,BGP,BMP2 and VEGF.CGRP can signifantly promote HUVECs proliferation ,migration ,tube fube formation,as well as the expression of CGRP recptors ,focal adhesion kinase ,VEGF, VEGF receptors 1,VEGF receptors 2. Implantation of vascular and sensory nerve bundles into tissue-engineered bone may enhance the osteogenesis of tissue-engineered bone in reconstruction of large bony defects and can also improve the neuropeptide contents and neuropeptide receptors.Implantation of vascular bundles into tissue-engineered bone could augment new bone prodution and capillary vessel formation with up-regulated expression of VEGF.SO, the neuropeptide can effect on the receptors of BMSCs and HUVECs,resulting in osteogenesis and angiogenesis and play an important role in tissue-engineered bone.
