中文摘要: 细菌的VI型分泌系统(T6SS)是一种新发现的大分子分泌系统,在多种病原细菌的致病性及环境适应性中发挥重要作用。本课题组从最近测序的水稻细菌性条斑病菌(Xoc)GX01菌株全基因组中注释了编码T6SS系统的同源基因。先导实验结果显示,注释的编码转位蛋白的基因hcp1突变导致该菌的致病力显著降低,提示T6SS可能与Xoc的致病功能相关。本研究的目的是阐明T6SS与Xoc致病性之间的关系,并鉴定依赖T6SS的效应子及基因。研究内容包括:1.构建T6SS的关键基因的缺失突变体,通过表型和致病力检测,明确T6SS在Xoc致病中的作用;2.构建T6SS操纵子表达gus报告子,筛选T6SS基因诱导表达培养基;3.进行野生型菌株与T6SS关键基因突变体间的分泌蛋白组比较,鉴定Xoc的T6SS效应子及基因。研究结果将为下一步鉴定T6SS效应子的寄主靶标、阐明T6SS在Xoc致病机制中的作用机理打下基础。

英文摘要: The bacterial type VI secretion system (hereafter T6SS) is a recently discovered macromolecular secretion system which plays an important role in the pathogenicity and environmental adaptation of a variety of phytopathogenic bacteria. Our research group has annotated the homologous genes for encoding T6SS from the recently sequenced genome of the rice bacterial leaf streak disease pathogen Xanthomonas oryzae pathovar oryzicola (hereafter Xoc) GX01 strain. The pilot experimental results showed that the mutation in the annotated translocation protein encoding gene hcp1 resulted in significantly reduction in virulence of Xoc, suggesting that the T6SS may be involved in the pathogenesis of this pathogen. The overall aims of this proposal are to elucidate the relation between T6SS and the pathogenicity of Xoc, and to identify the T6SS effectors and their genes. The major research contents include: 1. elucidation of the role of T6SS in the pathogenesis of Xoc by mutant phenotypic analysis and plant assays; 2. Selection of the induction medium for T6SS operons by using the gus reporter; 3. Identification of the T6SS effectors and their genes by comparative proteome profiling of the Xoc wild type and the T6SS key gene mutant.
