中文摘要: 卷须的作用是攀援。保护地栽培条件下,任卷须自由生长会造成株间、株与吊蔓绳之间相互缠绕和遮挡,不仅影响栽培群体的通风与透光,也给植株落蔓等田间管理带来困难;更重要的是它与同节位的雌花有强烈养分竞争,因此去除卷须能显著提高黄瓜产量。生产中为减少养分消耗,便于田间管理,常将卷须摘除,但这种做法势必导致生产成本加大,因此开展无卷须性状的基因挖掘与功能研究对于黄瓜高产省力化栽培极具理论和实际意义。本项目利用课题组创制的无卷须突变体为材料,在无卷须基因(td)初定位基础上,利用二代测序和图位克隆相结合的方法克隆基因;采用qPCR,RNA原位杂交、td序列多样性分析、电镜观察和遗传转化等技术验证td候选基因的功能。该研究结果不仅能为黄瓜无卷须育种奠定理论基础和提供可直接利用的目标基因,而且对于葫芦科其它植物卷须发生和发育调控机制研究也具有重要借鉴作用,同时也能为葫芦科植物的系统演化研究提供基础性资料。

英文摘要: The tendrils are thigmotropic coiling organs that aid in mechanical support of vines. However, under the protected cultivation conditions, tendrils not only has lost its proper role, but also adversely affect ventilation and penetrating light of planting group, and discommode field farming operations. Moreover, tendrils intensely compete with female flower in nutrient at the same node of cucumber,thus,the current practice in cucumber production is removal of tendrils by manpower. However, it will inevitably lead to increased of production costs, and therefore tendril-less will be great theoretical and practical significance for cucumber cultivation. The project is applied based on the needs of research and production status of cucumber in china. We has screened a tendrilless mutant(named csem323) by cucumber EMS mutant library. Combined with the findings of preliminary mapping of tendrilless gene (td) , We will clone this candidate gene based on next generation sequencing methods, validate and analysis its function using qPCR, RNA in situ hybridization, gene site diversity analysis, electron microscopy and genetic transformation. The results can not only break through bottlenecks of stem tendrils research and provide the theoretical basis for cucumber breeding and the target gene for directly used for the cucumber breeding, but also have an important reference for the study of cucurbits stem tendril formation and developmental regulation, and also provide basic data for the phylogeny of the cucurbits.
