中文摘要: 本项目拟发展一类基于自组装核酸量子点复合纳米材料的多元癌症诊疗一体化技术。通过核酸分子的生物矿化效应和自组装特性构建用于癌症诊疗一体化的近红外核酸量子点纳米复合材料。利用量子点的高强荧光特性、多色性以及核酸适配体的高特异性和高亲和性对具有不同分子表型的肿瘤细胞进行多元同步成像,同时利用量子点所携带的siRNA作为基因药物对不同肿瘤细胞进行针对性治疗,在此基础上实现针对个性化诊疗的多元癌症诊疗一体化技术。本项目将通过DNA自组装的方法将用于生物识别的核酸适配体分子和用于癌症治疗的siRNA分子与核酸量子点相复合,获得一系列具有高荧光光强、高稳定性、高特异性、高生物相容性以及高肿瘤穿透性的高性能核酸量子点纳米复合材料,在细胞和活体水平上对具有不同分子表型的癌细胞和肿瘤进行多元同步诊断与治疗,探索并建立针对复杂肿瘤体系的癌症诊疗一体化技术。

英文摘要: This project will develop a novel strategy for cancer theranostics based on a class of self-assembled DNA-templated quantum dot nanocomposite. The near-infrared DNA-templated quantum dot nanocomposite is constructed via the unique biomineralization and self-assembly capabilities of nucleic acids molecules. Herein cancer cells with various phenotypes could be simultaneously imaged using highly luminescent multicolor quantum dots functionalized with DNA aptamers that exhibit high affinity and selectivity toward cancer cells;Meanwhile, cancer cells could be selectively treated with siRNA-based anticancer drugs. A multiplexed cancer theranostics technology will be establishedtowards personalized medicine. In this project nucleic acids aptamers and siRNA molecules will be self-assembled onto the DNA-templated quantum dots to form the nanocomposite with strong photoluminescence, high photostability, high selectivity, high biocompatibility, and high tumor penetration capability. Simultaneous breast cancer diagnostics and therapy will be conducted on cell and live animal level, and a cancer theranostics technology will be established for complexed tumor systems.
