中文摘要: 利用肝细胞体外构建肝组织体为肝衰竭的治疗带来了希望,但如何在肝组织体内实现血管再生,仍然是组织工程研究面临的一个难题。申请者以基质空间结构介导内皮细胞形成血管化组织的研究为切入点,提出一种在肝组织体中实现血管再生的新方法。本研究拟构建微管状凝胶并作为"模具",将肝细胞和内皮细胞分别固定在"模具"的内部和外壁,经过培养、诱导和去凝胶化,形成管状的三维血管化类肝组织体。考察该体系中血管化组织的形态、结构及性能,分析内皮细胞层粘附、伸展、生长、成熟并形成血管化组织的过程机制,确定干预及维持该组织的条件因素,以期实现对肝细胞极性结构和功能的部分修复。研究结果将可为体外肝组织中血管再生提供一个新的思路,也可为其他组织中实现血管化提供参考依据。

英文摘要: Hepatic organoids, fabricated by the hepatocytes in vitro, provide a promising therapy for the Liver failure. However, the main challenge in this issue is the revascularization in the hepatic organoids in vitro. Based on the spatial structure of ECM mediating angiogenesis, we present a new method to achieve the angiogenesis in the hepatic organoids. We intend to fabricate a micro-fiber "template" composed of gel. And then the hepatocytes are embedded within the "template" and the endothelial cells are adhered to the surface of the "template", respectively. After the process of culturing, inducing and de-gelling,a tubular vascularized hepatic organoid is fabricated. The morphology, organization, construction and function of the vascular tissue within this system will be investigated. Based on the results, we will analyse the mechanism of cellular adhesion, elongation, proliferation and maturity, which lead to the formation of vascular tissue.We also need to figure out the factors, which intervene in and maintain this vascular tissue. Furthermore, by utilizing this system, the partial polarity and function of hepatocytes are expected to be restored. This study will provide a new method for the revascularization in the hepatic organoids in vitro. The results will also provide a scientific proof for the angiogenesis
