中文摘要: 生物活性物质在消化过程中胃部强酸的杀灭及酶解的共同作用下,其生物活性折损严重。开发和构建相应的保护体系,使其顺利到达肠道有效部位,对保证其生物利用率及其活性功效发挥具有现实意义。本研究拟采用天然生物材料大豆分离蛋白和乙二胺四乙酸二酐(EDTAD)为原料,通过EDTAD与肽链上赖氨酸胺基发生反应,将羧基引入蛋白分子链,生成改性大豆分离蛋白。在葡萄糖酸δ内酯的辅助下,进一步构建成食品级安全的pH响应微凝胶体系,从而将其作为载体实现活性物质的肠道靶向控释。通过对微凝胶的形貌特性分析,核壳结构分析、微凝胶内部和微凝胶之间的相互作用力分析、微凝胶的pH响应性分析,探讨pH响应微凝胶的构建机制,及其随环境pH 值变化的响应机理。研究进一步以乳铁蛋白为模型芯材,考察该凝胶材料对乳铁蛋白的负载能力,及不同pH条件下在体外的释放规律,通过小鼠动物模型分析微凝胶在体内对芯材的肠道靶向控释性能。

英文摘要: It is well known that most bioactive substances can't perform actions unless they remain unchanged through gastric strong acid environment and enzymatic hydrolysis. To ensure their functions and biological efficiency, protective carriers must be developed to convey the substances intact into intestine where they normally play their roles. The present research intends to acquire modified soy protein isolate (SPI) through conjugating the carboxyl into the SPI, by means of grafting ethylenediaminetetraacetic dianhydrid(EDTAD), which provides the carboxyl, onto the amino group in lysine of natural material SPI. Non-toxic and food grade pH sensitive microgels will be assembled by the modified SPI assisted with glucono-δ-lactone, to realize its intestine-targeted controlled release of the bioactive substances as a carrier. The assembling and pH sensitive mechanisms will be explored by analysis of morphologies, the core-shell structure of the gel particles, the internal and external interaction of the gel particles, and the pH sensitive performance. Further, the research constructs a model with lactoferrin as a corematerial to study microgel system's load captivity and release at different pH in vitro, and its controlled release performance in the intestinal passage of mice in vivo.
