中文摘要: 甲型H1N1流感病毒NA和M基因起源于禽源H1N1亚型猪流感病毒(SIV),其它基因起源于三源重组H1N2亚型SIV。在甲型H1N1流感爆发之前,禽源H1N1亚型和三源重组H1N2亚型SIV曾零星感染人类,但并未引起流感大流行,由此推测甲型H1N1流感病毒NA和M基因可能影响其复制、致病、传播能力等生物学特性。国内猪群中流行的禽源H1N1亚型SIV的NA和M基因(非甲型H1N1流感病毒NA和M基因,二者起源相同)对重组SIV的生物学特性有何影响,这是一个未知数。本研究以此为出发点,结合我们前期在国内猪群中分离到的禽源H1N1亚型和三源重组的H1N2亚型SIV,搭建三源重组H1N2亚型SIV的反向遗传操作技术平台,拯救含禽源H1N1亚型SIV的NA和M基因的重组SIV,研究禽源H1N1亚型SIV的NA和M基因对重组SIV生物学特性的影响,为解析SIV的复制、致病及传播机理奠定基础。

英文摘要: NA and M genes of 2009 pandemic H1N1 virus originated from the avian origin H1N1 subtype swine influenza virus, while other genes came from the triple reassortant H1N2 subtype swine influenza virus. Before the outbreak of 2009 pandemic H1N1 influenza, the avian origin H1N1 and triple reassortant H1N2 subtype swine influenza viruses sporadically infected humans, but they did not cause an influenza pandemic. So NA and M genes of 2009 pandemic H1N1 virus might affect its replication, pathogenicity, transmission, and other biological characteristics. Previously the avian origin H1N1 and triple reassortant H1N2 subtype swine influenza viruses have been isolated from pigs in China. In this study, the reverse genetics technology platform of the triple reassortant H1N2 subtype swine virus will be built firstly. Then a series of recombinant viruses containing the NA and M genes of avian origin H1N1 swine virus will be rescued. Finally the biological characteristics of these reassortant viruses, such as growth characteristics in vitro, plaque morphology, neuraminidase activity, virus morphology, replication and pathogenicity in mice, and the ability to transmit in guinea pigs, will be investigated. Our study will further explain the mechanism of swine influenza virus replication, pathogenesis, and transmission.
