中文摘要: 神经干细胞移植为重建损伤脊髓的连续性并恢复其功能提供了可能。如何调控移植的神经干细胞向神经元定向分化并促进新的神经纤维再生、促进细胞间建立新的联接是提高和修复脊髓功能的关键。EZH2是重要的表观遗传调控蛋白,参与调控干细胞自我更新和定向分化。神经干细胞在自我增殖及向不同终末细胞分化时EZH2表达水平不同。课题小组成员已观察到EZH2对周围神经元轴突再生速度有明显调节作用。本研究设计利用基因电转化技术将抑制基因或表达基因导入神经干细胞,调控其EZH2的表达水平;通过体内体外实验观察不同EZH2表达水平调控神经干细胞向神经元定向分化及促进神经纤维再生和新神经连接形成中的作用;利用荧光蛋白示踪技术及电生理和行为学检测观察脊髓损伤动物模型脊髓结构和功能恢复情况,探寻EZH2表观调控神经干细胞增殖分化的作用机制,证实表观遗传调控在脊髓损伤修复中的作用。为临床细胞学治疗脊髓损伤及相关疾病提供实验依据。

英文摘要: Neural stem cells transplantation for spinal cord injury to address pathological reduction of neurons and neuronal targets after the interruption. How to control differentiation of transplanted neural stem cells into neurons and nerve fiber regeneration, new targets for cells is the key to improving and repairing spinal cord function. Epigenetic regulatory mechanisms are central to maintain self-renewal and differentiation of stem cells biology. EZH2 is the epigenetic regulation of gene expression plays an important role in protein. Neural stem cells to self-when different terminal cell proliferation and differentiation, different levels of EZH2 expression. Studies by transgenic technology designed to control their genes into neural stem cells expression of EZH2, reach regulation of neural stem cells differentiation into neurons and promote the regeneration of nerve fibers and the purpose of the formation of new neural connections, using fluorescent protein electrophysiological and behavioral tests of tracer technique and observation of animal model of spinal cord structure and functional recovery of spinal cord injury. Explore EZH2 apparent mechanism regulating neural stem cell proliferation and differentiation, confirmed that epigenetic regulatory role in the injury and repair of the nervous system. These research ideal was a bold essay established in the former research foundation at home and abroad.
