中文摘要: 本课题在成功复制颅脑损伤模型的基础上,从多肽组学、分子生物学、电生理学水平研究针刺干预颅脑损伤的保护机制。颅脑损伤后取伤侧顶叶皮层组织,运用多肽电泳、荧光定量PCR、Western免疫印迹法、免疫组化法、细胞离子成像和膜片钳等多项现代技术研究颅脑损伤过程中的不同时段针刺对脑组织中的多肽、slit2 mRNA和蛋白表达、Nogo-AmRNA和蛋白表达、镁离子和钙通道的调制作用及相互关系,着重从多肽组学和效应信息转导系统的不同角度研究颅脑损伤的病理过程,深入探讨针刺对本病的作用机制,为针刺在颅脑损伤中的临床应用提供实验依据。本课题从宏观和微观的不同层次分析针刺对颅脑损伤的保护机制,以便发展针灸理论,推动针灸学国际化进程。

英文摘要: To study the expression of differential proteomics,differentially expressed genes, [Ca2+]i ,5-HT 、 NE、5-HT、NE、E、DA、5-HIAA、HVA、DOPAC in local organizations of free-fall model of traumatic brain injury rats by the ways of differential proteomics and Dynamic confocal laser scanning microscope and Fluorescent molecular probe techniques and gene chip technology; To study the effect of electro-acupuncture on histiocytic substances in rats brain ,provide the expeimental foundation for the early application of electro-acupunture on CCI.Not only 10 different expressed proteins and 3 genes such as Capn8,Sphk1 and Ccl2 were identified,but also the effect of the correct to the metabol- ic disorders of Neurotransmitter and acupunture to injuried organization [Ca2+]i were proved.Acupuncture does good effect on the CCI.These different proteins take part in scathe and repair in the way of energy metabolise, composition and hydrolization of protein, production and cleaning of free radical and reconstitution of frame of nerve cell of CCI rats.
