中文摘要: 本项目拟以牛乳主要过敏原α-乳白蛋白为研究对象,采用牛乳过敏患者的特异性IgE抗体和噬菌体随机肽库技术,亲和淘选构象性模拟肽,并通过网络工具对模拟肽进行鉴别;然后,借助生物信息学方法对模拟肽进行空间模拟转换成构象性表位。在此基础上,采用亲和力常数的测定和免疫竞争方法对构象性表位进行确证。最后,对构象性表位的氨基酸残基特性和表面结构特征进行分析。 通过本项目工作,将定位出牛乳α-乳白蛋白构象性表位,进一步阐明α-乳白蛋白致敏的物质基础,同时揭示牛乳α-乳白蛋白过敏原的结构识别特征,可望为今后基于表位靶标而制备低过敏乳制品提供致敏结构的关键信息。

英文摘要: With the aim of the major allergen bovine α-lactalbumin, mimotope peptides were derived from phage display peptide library by screening against the specific serum IgE from patients with milk allergy. Then the positive mimotope peptides distinguished by web tool were predetermined as the conformational epitopes through bioinformatics tool. On the basis of this, the conformational epitopes were verified by affinity constants and competitive immunoreactivity.The conformational epitopes were finally identified by characteristics of amino acid residues and surface structural features. The conformational epitopes on bovine α-lactalbumin were determined in the research, which would clarify the material basis for the allergenicity of α-lactalbumin and reveal the characterization of conformational epitopes.It is expected for providing the key information on the structure basis of food allergen in preparation of hypoallergenic dairy products on epitope-based target.
