中文摘要: 卡帕藻是生产卡拉胶的主要原料,但规模化养殖中环境胁迫导致的冰样腐烂病频发。前期研究中本课题组发现了异枝卡帕藻对逆境抗性高于长心卡帕藻的特殊现象,为探究产生该特殊现象的原因,本项目拟:1.比较逆境对异枝卡帕藻和长心卡帕藻光合原初反应和电子传递影响的异同;2.从光合作用关键蛋白和关键酶变化角度,比较逆境下两藻的光合系统II(PSII)的D1蛋白和光合系统I(PSI)的51kDa与45 kDa蛋白的表达量、碳同化关键酶Rubisco酶量及酶活,及其编码上述蛋白大亚基的基因(psbA、psaA和psaB、rbcL)表达量的变化;3.从活性氧代谢调控角度,测定逆境下两藻活性氧(O2-, H2O2)的含量及其淬灭酶系统(SOD,APX,CAT,MDAR,DHAR,GR,GPX)的酶活变化。通过上述研究,以期从光合作用和活性氧代谢调控角度综合阐明异枝卡帕藻的特殊抗逆机制,为卡帕藻的健康养殖提供理论指导。

英文摘要: Kappaphycus is the main raw material for the production of κ-carrageenan. However, ice-ice disease caused by enviromental stress burst frequently during the cultivation of the Kappaphycus. The special phenomenon that Kappaphycus striatum showed more resistance to enviromental stress than Kappaphycus alvarezii,was discovered by us in our former reasearch . In order to study the reason for the special phenomenon, the following works are planed to be accomplished. Firstly, differences in the status of the primary reaction of photosynthesis and electron transportation between K. striatum and K. alvarezii incubated in enviromental stress will be analyzed. Secondly, the quantity of PSII key protein (D1 protein) with the encoding gene psbA, the quantity of PSI key proteins (51kDa and 45 kDa proteins) with the encoding genes psaA and psaB and the quantity and activity of Calvin cycle key enzyme (Rubsico enzyme) with large-subunit encoding gene rbcL will also be studyed. Thirdly, the quantity of active oxygen key products (O2o-, H2O2)and activity of quenched enzymes (SOD, APX, CAT,MDAR,DHAR,GR,GPX) will be detected when the K. striatum and K. alvarezii are incubated in the same stress condition.
