中文摘要: 多环萜类天然产物以其重要的生物活性(如抗癌、抗菌和鞘氨醇激酶抑制剂)、独特的化学结构和潜在的应用前景使其成为一个备受关注的研究领域。本项目在已有工作的基础上,运用有机合成化学新概念、新知识和新技术,根据目标分子的结构进行巧妙设计,发展高效合成策略,力争在国际上率先完成一系列具有重要生物活性的多环萜类天然产物的不对称全合成。在项目实施过程中,结合项目组报道的串联反应和现代有机合成方法学中的新型串联反应等来设计尽可能短的合成路线,符合"原子经济性"和"步骤经济性"理念,同时也考虑所选择串联反应能否满足多样性合成的需要。项目组通过多环萜类天然产物全合成带动相关有机合成方法学的发展,而发展的有机合成方法学反过来更好地为更多天然产物全合成服务。通过本项目的实施为萜类天然产物全合成和相关有机合成方法学领域的理论创新做出一定贡献。

英文摘要: Polycyclic triterpenoidal natural products have attracted increasing attention thanks to their important biological activities, such as anticancer, antibacterial and sphingosine kinase inhibitor, as well as their unique chemical structures and potential application prospects. In connection with our ongoing work in this field, herein we would try our best to accomplish several first asymmetric total syntheses of bioactive polycyclic triterpenoidal natural products using efficient synthesis strategies. To achieve this goal, we would like to study their unique chemical structures, and then to design desired synthetic routes by using the new concepts, new knowledge and new technologies in the field of synthetic organic chemistry. To achieve the concepts of "atomic-economy" and "step-economy", we would like to design the shortest possible synthetic routes by using novel tandem reactions in modern organic synthesis along with useful tandem reaction reported by us. Moreover, we would like to pay attention to whether the tandem reactions can meet the requirements of diversity-oriented synthesis. We would like to develop new synthetic methodologies during our total synthesis, and, in turn, to improve our total synthesis by using new-developed synthetic methodologies.
