中文摘要: 我们从中药材川芎中分离出的活性成分Z-藁本内酯能够保护水稻免受精异丙甲草胺的药害,但是其具体作用机理尚不明确。我们假设Z-藁本内酯主要作用机制是通过提高GSTs活性促进水稻体内精异丙甲草胺的代谢;同时推测Z-藁本内酯对水稻体内精异丙甲草胺吸收传导的影响可能是Z-藁本内酯作用机制的一部分。采用同位素标记、HPLC、LC-MS、RT-qPCR、RNAi、过量表达、转基因和生物测定等技术方法,研究Z-藁本内酯对水稻体内14C-精异丙甲草胺吸收传导的影响;同时,对水稻体内14C-精异丙甲草胺的代谢产物变化进行研究,并研究代谢调控基因GSTs(tau、phi和lambda)的差异化表达;进一步对有差异化表达的基因进行过量表达和RNA干扰,确定关键调控基因。目的就是为了确定Z-藁本内酯保护水稻免受精异丙甲草胺的药害作用机理,为此类新型安全剂的开发应用提供理论指导,具有重要的基础研究价值。

英文摘要: We found Z-ligustilide separated from Ligusticum chuanxiong hort could protect rice from S-metolachlor injury, though its mode of action is not known. We presume Z-ligustilide accelerate the metabolism of S-metolachlor through the increased GSTs-catalyzed reaction, whilst the effect of Z-ligustilide on uptake and translocation in rice seedlings maybe play a certain role in its mode of action. To understand its mode of action, we will research uptake, translocation and metabolism of the 14C-labelled S-metolachlor in rice seedlings using methods of isotope labelling, HPLC and LC-MS. Subsequently RT-qPCR will be taken to detect the differential expression of metabolism-related GSTs (tau,phi and lambda), and then the overexpression and RNAi of differentially expressed GSTs genes in transgenic rice is an attempt to verify their effect on S-metolachlor metabolism. Our study is a valuable fundamental research could provide theoretical guidance for the development and application of a new type of safener like Z-ligustilide.
