中文摘要: 细胞分裂素在植物生长和发育过程中起重要的作用,但对于细胞分裂素调控植物生长发育的分子机理知之甚少,特别是在重要的粮食作物水稻中几乎为空白,至今还没有水稻细胞分裂信号途径突变体的分离鉴定及其相应基因克隆的报道。本研究通过筛选水稻突变体库,得到一个细胞分裂素不敏感突变体Oscki1,遗传分析表明该突变性状受一对隐性单基因控制,本项目将采用图位克隆法克隆OsCKI1基因,通过遗传互补、超表达、RNAi等技术验证其功能,再利用亚细胞定位、启动子融合GUS报告基因、组织切片、生理生化分析等手段阐明OsCKI1基因的表达位点和作用模式,为进一步揭示水稻细胞分裂素信号转导的分子机制提供理论依据。

英文摘要: Cytokinins play important roles in plant growth and development, however, how cytokinin regulate plant growth and development is still largely unknown, especially in terms of the important crop rice.Until now there is no report about the identification and genetic cloning of mutants of cytokinin signalling pathway in rice.Our research indentified a cytokinin-insensitive mutant Oscki1 from a rice mutant library. Genetic analysis revealed that the phenotype was controled by a single pair of recessive gene. In this study, OsCKI1 will be cloned by map-based cloning and characterized by genetic complementation, overexpression and RNA interference. Its expression pattern and function will be further clarified by subcellular localization study, promoter-GUS fusion reporter assay, histological,physiological and biochemical analysis. It will provide theoretical basis for further illumination of the molecular mechanism of cytokinin signaling transduction pathway in rice.
