中文摘要: Nell-1是一新发现的具有较强成骨活性的成骨基因。为了研究NELL1是否能有效防治磨屑所致的骨溶解,本实验通过构建重组腺病毒Ad-GFP-NELL1将NELL1基因转染到细胞(包括骨髓间充质干细胞和成骨细胞)及动物模型体内(包括大鼠牵张成骨及小鼠颅骨骨溶解模型),应用Micro-CT观察骨再生,并对新生骨进行生物力学性能的检测和组织病理观察。本研究证明了构建成功的重组NELL1基因能有效促进骨髓干细胞及成骨细胞的成骨分化作用;在骨缺损及骨溶解动物模型中,NELL1能促进高质量的骨再生,平衡骨溶解所致的骨量丢失,为防止人工关节磨屑造成的骨溶解,治疗人工关节假体松动提供新的治疗手段。

英文摘要: We investigated the therapeutic effects of a craniosystosis-associated molecule, NEL-like molecule-1 (NELL1; NEL [a protein strongly expressed in neural tissue encoding epidermal growth factor like domain]), on osteolysis induced by polyethylene (PE)-particle debris.We used cells culture(BMMCs and osteoblasts) and animal models (calvarial osteolysis models and distraction osteogenesis models)with adenovirus (Ad)-mediated gene transfer,which were analysized by 3-D micro-computed tomography (micro-CT), real-time PCR, histology and biomechanical testing.We found that Ad-GFP-NELL1 could induce BMMCs and osteoblasts' ossification,promote high-quality bone regeneration to compensate the osteolysis in vivo, and could be a new treatment for osteolysis by promoting bone regeneration.
