中文摘要: 昆虫变异眼色作为典型的遗传标记,具有重大的理论研究价值和很大的应用潜力。本项目组在前期成功选育褐飞虱红眼纯合品系和开展遗传学研究的基础上,拟以褐眼野生型和红眼突变型为研究对象,通过不同色素前体物添加对突变眼色的修复性试验明确红眼突变的生化基础,定位引起眼色突变的眼黄素合成/运输环节(代谢环节);利用生物信息学和分子生物学技术,克隆代谢环节中的眼黄素相关基因,并在野生型和突变型中进行对比,确定突变基因和突变方式;利用mRNA定量技术和蛋白质体外表达技术,分析突变基因量变和所编码蛋白生化特性的改变;利用RNAi技术对野生型眼色的干扰性试验验证突变基因的功能。项目预期将揭示褐飞虱红眼突变的生化基础和分子机理。利用褐飞虱眼色突变基因作为遗传标记,不仅可以促进褐飞虱功能基因组学和蛋白质组学的研究,还可以促进与红眼突变相关的其他生物学性状的研究。

英文摘要: As typical genetic marker, insect mutant eye color has significant research value and high potential for use. In our laboratory, a homozygous colony of red-eyed Nilaparvata lugens St?l (brown planthopper, BPH) was established. On the basis of previous genetic research, we will reveal the mutant mechanism of red eye color in BPH based on the comparison beween the wild-type and mutant-type colonies. First, locate the mutation step in xanthommatin biosynthesis/transport (metabolism step) by the mutant-type rescue experiment. Second, acquire xanthommatin-related genes in the metabolism step using the biological information and molecular biology technology, and ascertain the mutant gene and mutant fashion by sequence analaysis. Third, compare the differences in the mRNA level and the biochemical characteristics of recombinant protein of the mutation gene using real-time PCR and heterologous gene expression technology. Then, verify the mutant gene function by interruptting the wild-type eye color with RNAi technology. This study aim to reveal the biochemical basis and molecular mechanisms of red-eyed mutant in BPH. Using the mutant eye color gene as a visible genetic marker, it will not only improve the studies on functional genomics and proteomics in BPH, but also accelerate the studies of BPH biology traits related
