中文摘要: 基于前一个国家基金项目的研究,本研究将继续瞄向"通过细菌生物膜自然属性的探讨,解析生防细菌抗菌活性调控机制"的主题,以具有重要生防特性的解淀粉芽孢杆菌为对象,针对新发现的"生物膜表型异质性变异伴随抗菌活性衰减"的客观现象,建立细菌动态变异的试验体系,揭示生物膜表型异质性变异的动态过程及其生态对策;采用转录组测序从整体上分析生物膜表型异质性变异的调控机制;在此基础上,重点聚焦DegS-DegU、SinR/sinI等调控系统,围绕"运动性"、"芽孢形成"和"抗菌活性"事件,采用关键基因突变以及实时定量PCR技术,探讨不同变异表型下细菌对这些事件的调控特征,分析事件之间分子调控通路上的关联,从表型和分子性状上,揭示生物膜表型异质性与抗菌活性的相关性,进而发现解淀粉芽孢杆菌抗菌活性调控的重要机制。本研究对探索细菌生物膜形成机制、揭示细菌抗菌活性的调控机制,具有重要科学意义和潜在应用价值。

英文摘要: The project is brought forward based on the progress on the study of previous project of the National Natural Science Foundation of China, and on the extensive review on the domestic and international studies. The study will continue to aim at the regulation mechanism of antimicrobial activity of Bacillus amyloliquefaciens using the natural quality of bacterial biofilm. In the project, phenotype heterogeneity of bacterial biofim will be used as the object to reveal the regulation mechanism of the phenotype heterogeneity variation and therefor influencing the antimicrobial activity. To achieve that, we will establish the experimental systems of the bacterial dynamic mutation to reveal the dynamic progress of the mutation and ecological characteristics of adaptive variation. The methods of RNA-seq, genic mutation and Real time PCR will be used to reveal the regulation mechanism of the phenotype heterogeneity variation of the bacterial bioflm. "DegS-DegU" and "SinR/sinI" will be focused on and Bacterial motility, Bacterial spore formation and Antimicrobial activity will be used as the key events to discuss the relationship between phenotype heterogeneity of bacterial biofim and antimicrobial activity of B. amyloliquefaciens.
