中文摘要: 随着对RAS系统研究的不断深入,人们发现局部RAS的存在对组织的生理功能和结构起着非常重要的作用,其中对男性生殖功能的调节作用研究甚少。在本次申请者参与的前期自然基金项目中发现AT1基因敲除(AT1KO)可致雄性小鼠不育而雌鼠正常。根据RAS各组分及相关调控酶在雄性生殖系统中的大量分布,提出RAS可能是继下丘脑-垂体-性腺轴之后又一"肾主生殖"的理论实质。并据此理论,试图以补肾填精法来调整RAS系统,从而改善男性生殖功能。 课题借助已成功的AT1KO雄性小鼠,运用生化、免疫组化、分子生物学等对血浆中循环和睾丸中局部的RAS组分、性激素水平进行检测,阐明循环及局部RAS对男性生殖系统的影响;并以五子衍宗丸进行干预,观察循环和局部RAS、性激素水平,睾丸组织形态、精子活力等,阐明补肾填精法改善由RAS敲除导至不育的机理及疗效。

英文摘要: With the deep research of RAS system,the local RAS was found to be very important in physiological function and structure of the organization.The researth of the male reproductive function is very little among it. In the earlier NSFS we found that AT1KO can cause infertility in mice,but normal in females. According the Large distribution of the various components and related enzyme in RAS,we think RAS may be another thoery essence of "kidney controlling reproduction" after the axle of hypothalamic-pituitary-gonadaxis. We try to regulate the RAS by nourishing kidney in order to improve the male reproductive function. Depend on the secceeded AT1KO mice,we will detect the components of RAS in cycling and the local and sex hormones by using the motheds of biochemistry、SABC and MICM,in order to clarify the differents of RAS in cycling and the local.We also use Wuziyanzong Pills to research the RAS in cycling and the local 、Sex hormones、morphology of Testicular tissue and sperm motility,in order to clarify the mechanism and effect of improving the biologicalreproductivefunction of AT1KO mice by nurishing kidney.
