中文摘要: 本研究拟采用海带"860"自交系及长海带为亲本,杂交获得F1代,然后以"860"自交系为轮回亲本,通过连续回交,构建海带的近等位基因系遗传群体(NILs),从该群体的成熟个体中,采用表型和分子标记选择含有长海带基因组片段的目标个体,分离其雌、雄配子体进行培养保存,从而可将建立的海带近等位遗传群体(NILs)永久保存下来,获得海带的永久遗传群体,为海带数量遗传学的可持续性和重复性研究奠定基础,并为海带的良种选育提供材料基础;通过对海带BAC克隆的测序分析,获得批量微卫星标记,SNP标记,用以构建以共显性标记为骨干标记的海带遗传连锁图谱,对海带叶片长度进行QTL比较定位,借助近等位基因系群体(NILs),结合大样本分析,对叶片长度主效QTL区域进行精细作图。

英文摘要: In this research, a NILs (Near-isogenic lines) population being planned to be developed from the hybridization of recurrent parent "860" inbred line of Laminaria japonica and donor Laminaria longissima, will be used to fine mapping of available QTL for Laminaria blade length. Through isolating and culturing the gametophytes of marker selected individuals containing the part of genome of Laminaria longissima in the NILs population, the Laminaria NILs population can be permanently preserved for continuing and repeating research for Laminaria quantititaive genetics and Laminaria breeding in future days. SSR and SNP molecular markers, can be obtained by sequencing BAC clones. One genetic linkage map can be constructed using the codomainant markers, which can be used to compare with the available genetic linkage map mainly constructed with AFLP markers. With the Laminaria NILs population being developed in this research, fine mapping of the available QTL of Laminaria blade length, and cloning the relative genes, will be made possiblly.
