中文摘要: 抗病基因使植物从抗病性中获利,然而抗病基因的适应性代价假说提出了在无相应病原体时,抗病基因植株与对应的感病植株比需要消耗额外的能量与物质或产生负调节作用,导致植物适应性降低。这个假说已在插入/缺失型抗病基因中得到证明,但植物大多数抗病基因是非插入/缺失型,其是否也存在适应性代价是个重要的科学问题,目前已有的研究没有合理的思路和技术,因而没有得到证明。本项目选择拟南芥抗病基因Rps2不同程度的抗、感病生态型,利用基因定点整合技术构建遗传背景完全一致、不同个体在DNA同一位点含不同的单拷贝抗病的等位基因系,并通过统计这些基因系植株在无病原体环境中的一系列生物量指标,用实验证明非插入/缺失型抗病基因是否存在适应性代价及在不同程度感抗病群体中的变化规律。同时,选择水稻的抗稻瘟病Pita基因作平行实验,以检验非插入/缺失型抗病基因的适应性代价是否受植物种类、抗病基因种类的影响。

英文摘要: Resistance genes protect plants against a wide spectrum of diseases; however, it can also entail costs due to the allocation of resources or toxicity of defensive products. The hypothesis of fitness cost of resistant genes has been proven in the insertion/deletion resistant genes. Based on the fact of the majority of plant disease resistance genes are non-insertion/deletion type, whether fitness cost exist in this type of resistant genes is considered to be the most representative for general. All of the current technologies are not suitable to prove this issue. So this proposal plans to select different donors of resistant gene Rps2 in Arabidopsis thaliana according to different disease symptoms, through site-specific recombination system, the isogenic lines of the same genetic background containing series of allelic genes in the same insertion site on chromosome would be constructed; by recoding and statistical analysis of fitness related traits between each pair of isogenic lines, we try to prove that the fitness cost would be detected in non-insertion/deletion resistant genes and unravel the variation in different isogenic lines. Meanwhile, the choice of rice blast resistant gene Pita for parallel experiments would pave the way to know effect of the type of plant on fitness cost.
