中文摘要: 在本实验室的前期工作中,我们构建了一系列与精子发生相关的蛋白表达谱系,并筛选到了一批蛋白质,本研究选择Symplekin蛋白开展深入的功能研究。在体细胞的经典多聚腺苷酸化通路中,Sympk蛋白作为一个脚手架协调CPSF和CstF的相互作用,而生精细胞基则通常是利用非经典但特异的多聚腺苷酸化信号通路,因此在其高表达的精子发生过程中,Sympk蛋白是否参与生精细胞特异的多聚腺苷酸化通路,是否依然作为脚手架协调相关蛋白的相互作用还尚未阐明及验证。本研究将通过制备Sympk生精细胞特异性条件敲除小鼠模型深入分析其在精子发生特定多聚腺苷酸化通路中的作用,并试图能够阐明参与此通路的Sympk相关的蛋白复合体。

英文摘要: We have constructed a series of proteome profiles describing spermatogenesis and this large-scale identification of proteins provides a rich resource for elucidating the mechanisms underlying male fertility and infertility. Here we focused on Symplekin to explore detailed function. In somatic cells, sympk regulates the interaction of CPSF and CstF as a scaffold protein in the classical pathway of polyadenylation. However in the spermatogenesis, spermatogenic cell usually use non-classical but specific polyadenylation pathway. So during spermatogenesis, we still do not know whether sympk participates in the specific polyadenylation pathway of spermatogenic cells. This study will explore the function of Sympk during polyadenylation of spermatogenesis through the conditional Sympk knock out mice model and try to clarify the protein complex associated with sympk.
