中文摘要: CDK(Cyclin Dependent Kinase)属于丝氨酸/苏氨酸蛋白激酶家族,其生物学功能主要体现在调控细胞生长周期,此外还在RNA转录与剪切,细胞分化等过程中发挥作用。通常不同的CDK要和相应的Cyclin结合来获得生物活性。 根据与动物CDK11蛋白质序列的同源性分析以及CDK11和CyclinL结合的保守序列PI/LTSLRE,在拟南芥中分离鉴定了CDKG1和CDKG2。前期研究结果表明,CDKG1缺失突变可以使拟南芥的花粉育性降低甚至缺失,育性的降低可能与花粉母细胞的减数分裂异常相关,但是其调控机理目前还不清楚,可能与小麦Ph1 CDKs功能相关,涉及目前未知的CDK/cyclin复合体对染色体配对、联会和重组的调控机制。本项目拟利用相关突变体,对CDKG参与植物花粉母细胞减数分裂调控机理及调控网络开展研究,为基于有性生殖与基因重组交换调控的作物遗传改良提供理论依据。

英文摘要: CDKs belong to a protein family of serine/ threonine protein kinases that, with specific cyclins, regulate many biological processes including cell cycle, RNA splicing and transcription regulating. The CDKG1 and CDKG2 proteins from Arabidospsis are predicted to contain a conserved cyclin binding domain " PLTSLRE". CDKG kinases are structurally related to the Ph1-related kinases in wheat implicated in controlling homeologous chromosome pairing. CDKG1 has been shown to interact with cyclin L but no function has been ascribed to either partner. We show that CDKG1 is essential for successful male meiosis when plants are grown at high temperature . Cytological analysis of a T-DNA insertional mutant in the CDKG1 gene reveals that the fertility failure is due to defective meiosis.Bivalent formation in meiotic prophase I is much reduced leading to random chromosome distribution and formation of abnormal meiotic products. However,how CDKG1 regulate meiosis proress in pollen mother cells is still unclear. In this project, we will try to investigate the mechanism of CDKG1's function during meiosis and the gene network which involed in this progress. The main goal is to provide a theoretical foundation for transfering desirable agronomic traits into crops from wild relatives.
