中文摘要: 关节软骨的病变常伴随软骨下骨的缺损,因此骨软骨一体化修复一直是再生医学和骨科面临的困难之一。传统的骨软骨组织工程支架往往忽略骨—软骨界面这一重要结构的构建,难以实现其一体化修复;同时,如何同步介导干细胞在支架不同相的特异性梯度分化,实现与天然骨软骨组织在成分与结构上匹配也是制约骨软骨一体化修复临床应用的难题。本课题利用微流体器件构建具有羟基磷灰石梯度分布的水凝胶材料,以成分仿生骨软骨;然后将其负载干细胞,并置于由成骨及成软骨诱导液形成溶液梯度的微流体器件中,通过调控两种诱导液在水凝胶中的梯度分布,将不同区域的干细胞分化为成骨细胞或软骨细胞,从而制备由骨相到软骨相,具有连续过渡结构的骨软骨修复材料。本课题围绕骨软骨一体化修复与重建两大难题,应用微流体与组织工程相结合的新方法,构建可介导干细胞梯度分化的高度仿生骨软骨修复材料,期望通过不同技术领域的交叉突破骨软骨组织工程一体化修复的瓶颈。

英文摘要: Injury to articular cartilage, affects both articular cartilage and the underlying subchondral bone, remains a significant challenge in orthopedic surgery. Current osteochondral tissue engineering not only neglects the biomimic interfaces between generated bone and cartilage zones, but also cannot simultaneously steer osteo/chondrogenesis of stem cells in one scaffold. In this project, we will develop an innovative method which fuses the fields of microfluidics, biomaterials and tissue engineering to design a biomimic materials for osteochondrogenesis. To mimic the compositions and cell distribution of osteochondral tissue, we will design a stem cell-laden bioactive hydrogel materials with hydroxyapatite and osteo/chondrogenic differentiated medium gradients generated by microfluidic devices. The format using gradient-generating device would be outstanding superiorities in stem cell differentiation and interface generation. The execution of this project will not only provide a feasible and promising way for articular cartilage regeneration, but also may open a new door for tissue engineering.
