中文摘要: 对韧带缺损的修复是目前临床上急需解决的问题。当前使用的韧带缺损修复替代物均有不可避免的缺点。将韧带/肌腱经过脱细胞处理得到的脱细胞胶原纤维具有天然韧带的形态结构及复杂的生物信号,最有希望成为理想的替代物。但由于脱细胞过程去污剂的使用,降低了该材料的生物力学性能,且影响其在体内重建。课题组前期将牛肌腱低压冻干,常温下打开腱膜并分离得到肌腱纤维束,其力学性能未受影响。且细胞成分无腱膜包绕,脱细胞过程大大简化,有望不使用去污剂,最大限度保留肌腱纤维固有结构、力学强度及复杂的生物信号。本课题在前期研究基础上,进一步研究不含去污剂的脱细胞方法,对获得的脱细胞胶原纤维材料的生物学特性进行分析,包括组织学形态、成分、生物力学、组织相容性等。继而将该材料用于韧带缺损模型的修复,并通过组织学、生物力学等检测对修复效果及体内韧带结构的重建进行评价。本研究对新型脱细胞胶原纤维材料用于韧带缺损的修复重建至关重要。

英文摘要: The repair of ligament coloboma is a problem that requires urgent solutions in clinical practices. Almost all currently adopted substitutes for ligaments have weakness in some aspects. A very promising and ideal substitute is acellular collagen fiber obtained by decellularization of ligaments or tendons , which possesses the structure of natural ligaments and can carry complex biological signals. But the use of decontaminant in the procedure of decellularization adversely reduces the mechanical properties of acellular collagen fiber and impairs its reconstruction in vivo. In our previous research, we have presented a method of obtaining acellular collagen fiber to guarantee the mechanical properties are not affected, during which bovine tendons are first lyophilized and then the tendon fiber bundles are obtained by opening the aponeuroses and separation in normal temperature. Another advantage of our approach is that the cells are not surrounded by the aponeuroses and the procedure of decellularization is greatly simplified, which will probably open the opportunity to give up the use of decontaminant to maintain the original structure of tendon fibers, the mechanical properties and the complex biological signals.
