中文摘要: 基于不对称有机小分子催化的串联反应合成天然产物和药物分子是目前有机化学发展的难点和热点。不对称有机小分子催化是近年来发展起来的构建手性化合物的新方法,尤其是有机小分子催化的串联反应在构筑含有多手性中心环状化合物时,表现出了其他方法无法比拟的优势。将这类方法应用于全合成时,更是将绿色、经济和高效的理念引入全合成中。本项目将在此领域展开深入的研究,选取了三个具有药用价值的天然产物Lycopladine A,Peribysin E和Aphanorphine。通过采用不对称有机小分子催化的串联反应,快速构建它们的骨架,并引入手性中心,同时引入必要的官能团。并以这种高效的方法为基础,实现目标分子高效简洁的全合成

英文摘要: Total synthesis of natural products and drugs based on asymmetric organocatalysis cascade reactions has been at the forefront of organic chemistry. Over the last decades, asymmetric organocatalysis have been developed a new method for the construction of chiral compounds, especially asymmetric organocatalysis cascade reactions in constructing multiple chiral centers cyclic compounds, showed that other methods cannot match advantage. It will bring new opportunities for total synthesis. This project will conduct in-depth research in this emerging field. We selected four natural products with potential medicinal value, Lycopladine A, Aphanorphine and Peribysin E. Through asymmetric organocatalysis cascade reactions, their skeletons can be rapidly constructed, chiral centers can be formed and the necessary functional groups can be introduced. Based on this efficient method, realize and concise total synthesis of the target molecules.
