中文摘要: 本研究以番茄花柄为试材,首先明确器官脱落过程中外植体垂直方向和水平方向上的生长素浓度梯度,并根据PIN时空表达,蛋白含量以及定位的变化规律,初步建立起调控脱落的生长素浓度分布及运输机制;其次,通过转基因技术沉默和超表达候选基因结合其对PIN蛋白定位的影响,筛选出调控脱落的关键基因;第三根据钙延缓脱落过程中,钙素对PIN相应基因表达及定位的作用,结合前期的研究结果,阐明钙素的调控机理并明确钙直接调控的相关成员;第四根据受钙调控的PIN蛋白,通过酵母双杂交技术筛选出特异调控PIN磷酸化的钙结合蛋白,找到钙直接作用的位点。最终结合以上结论,揭示钙在IAA延缓番茄花柄外植体脱落过程中的作用机制,为钙调控IAA信号作用模式研究打下基础。

英文摘要: In this study, tomato flower pedicel was used as experimental material. First, identify the auxin change of the explants vertically and horizontally in explants during the abscission, and build the auxin concentration distribution and transportation mechanism for regulating the abscission initially according to PIN Spatial-temporal expression, protein content and positioning change pattern; second, with the combination of RNAi silencing and over-expression candidate gene along with its affect on PIN protein positioning, screened out and identify critical genes for regulating the abscission; third, elucidate the calcium regulation mechanism and identify related members regulated directly by the calcium according to the effect of the calcium on corresponding gene expression and positioning of PIN; fourth, identified calcium-binding protein which regulates PIN phosphorylation rely on the yeast two-hybrid system and find the point on which the calcium acts directly. Finally, based on above conclusion, reveal the effect of the calcium on auxin inhibit abscission of the tomato flower pedicel, and make the foundation for the mechanism of calcium regulating IAA signal.
