中文摘要: G蛋白偶联受体(GPCR)是一类具有七跨膜螺旋结构的膜受体蛋白,组成人体内最大的蛋白质家族,主要介导细胞信号转导,是极其重要的药物靶点。趋化因子受体CXCR4属于GPCR家族,是人免疫缺陷病毒的一种共受体,可协助病毒感染人体细胞;此外,CXCR4在多种癌细胞表面大量表达,介导癌细胞的迁移。本项目利用昆虫细胞杆状病毒表达系统高量表达人源CXCR4蛋白,与多种多肽配体结合后,通过蛋白纯化和结晶条件筛选获得高质量的CXCR4与多肽配体的复合物晶体,收集X-射线衍射数据,最终解析其高分辨率晶体结构。通过以上研究,将进一步深入理解CXCR4与配体的结合模式,揭示CXCR4识别细胞外信号的分子机制,并为抗艾滋病和抗癌症新药研究提供分子结构基础。

英文摘要: G protein-coupled receptors (GPCRs), containing seven transmembrane helixes, comprise the largest protein family in human genome. The receptors sense molecules outside the cell and activate inside signal transduction pathways. GPCRs are involved in many human diseases, and represent the target of approximately 40% of all modern medicinal drugs. Chemokine receptor CXCR4, a member of GPCRs, acts as a co-receptor of Human Immunodeficiency Virus (HIV), which facilitates the viral entry. CXCR4 is over-expressed on the surface of many kinds of cancer cells, and mediates cancer metastasis. In this project, CXCR4 protein will be expressed using insect cell-baculovirus expression system. The peptide ligand bound CXCR4 protein will be purified and crystallized. Crystal structures of the receptor-peptide complexes will be solved by collecting high resolution X-ray diffraction data. The structural studies of CXCR4-peptide ligand complexes will deepen our understanding of CXCR4-ligand binding mode and cell signal recognization mechanism, and lay a foundation for carrying out structure-based drug discovery aimed at inhibiting HIV cell entry and cancer metastasis.
