中文摘要: 三七叶总皂苷PnGL为五加科植物三七茎叶的提取物,具有益气安神、活血止痛的功效,是传统中成药七叶神安片的主要原料。本项目前期的药理研究表明:PnGL具有明显的抗抑郁作用,其抗抑郁机制可能与五羟色胺能系统、去甲肾上腺素能系统以及多巴胺能系统等三种神经递质系统有关。我们将在此基础上,以中医药多成分、多靶点和多途径协同作用整体观的理念为指导,以抗抑郁活性评价为导向,系统研究PnGL的物质基础,确定抗抑郁活性组分或成分,研究主要活性成分的抗抑郁作用机制及它们之间可能的协同作用,以及构效和量效关系。根据抑郁作用机制及相关药理靶点的结构生物学信息,应用放射免疫学、免疫组织化学、大鼠慢性应激抑郁模型,以及各种光谱学技术和先进的生物物理研究方法,阐明PnGL对神经递质、神经细胞(因子)、神经信号转导通道的影响及相关分子机制。为开发新型抗抑郁中药新药提供理论和实验依据。

英文摘要: Total saponins from the leaves of Panax Notoginseng(PnGL) is the extract from the herb of Panax notoginseng (Burk.) F.H.Chen, araliaceae. PnGL can reinforce Qi for tranquilization and activate blood for relieving pain. It is the main material for the traditional Chinese drug Qiyeshenan Tablet. Our previous pharmacological study on PnGL has shown the facts: PnGL has an evident antidepressant effect depending on its close relationship with serotonin system, noradrenergic system and dopamine system. Basing on the previous study and the traditional chinese medicinal theories, such as the complexity of drug components and multiple targets of drug actions, further research needs to be carried out. The correlation of chemical compounds with antidepressant effect of PnGL should be observed with the method of bioassay-guided fractionation and isolation. The bioactive fractions or constituents in PnGL should be known. The antidepressant mechanism, the possible coefficient efficacy, the structure-activity relationship and the dose-effect manner between bioactive constituents should be researched. According to the present information of antidepressant mechanism and targets of drug actions, the effect of PnGL on neurotransmitter-nerve cell (neurotrophic factor)-neuronic signal transduction pathway should be investigated with
