中文摘要: 桑树是一种多用途的重要经济树种。桑树青枯病是一种威胁桑树生产的毁灭性细菌病害,影响桑树相关多个产业的发展。TGA是一类与植物生物胁迫抗性相关的转录因子,在水杨酸介导的植物系统获得抗性(SAR)信号转导途径中起重要调节作用。申请者前期工作表明:MaTGAs基因在桑树强抗品种中被青枯菌特异诱导表达,可能与桑树青枯病抗性密切相关。在此基础上,本研究拟进一步分析MaTGAs基因在青枯菌处理下的时空表达特性、蛋白产物的生化特性;同时通过MaTGAs沉默桑树和过表达模式植物鉴定其抗青枯病功能;并采用高通量测序技术比较分析转化株和对照株的转录组差异,以阐明MaTGAs的基因网络调控模式。该项目的实施有望在理论上阐明MaTGAs在桑树中的抗青枯病分子机制,为培育不同用途的桑树抗病新品种提供基因储备和理论依据。

英文摘要: Mulberry is an important multipurpose economic woody plant. Mulberry Bacterial wilt caused by Ralstonia solanacearum is a destructive bacterial disease, which affects the development of several industries related with mulberry. TGA is a group of transcription factors related with plant biotic stress resistance, and plays an important role in salicylic acid-mediated plant system acquired resistance (SAR) signal transduction pathway. Our preliminary works showed that: The MaTGAs genes were specifically induced by Ralstonia solanacearum in resistant mulberry varieties, which may be closely related to the resistance of mulberry Bacterial wilt. On this basis, in this study, we intend to further analyze the spatial and temporal expression characteristics on Ralstonia solanacearum treatment of MaTGAs, and the biochemical properties of their proteins. Then we will identify disease resistance function of MaTGAs by the methods of VIGS and overexpression in model plant, and analysis of the difference between the transcriptome of transformed and control plants using high-throughput sequencing technology, in order to explore the gene regulation network of MaTGAs. Through this study, it is expected to clarify the molecular mechanism of MaTGAs in Bacterial wilt resistance of mulberry in theory, and to provide gene pool and theoretical basis for breeding disease resistant varieties for different uses.
