中文摘要: 药对是中药复方配伍的一种特殊而相对简单的形式,很多中药复方都包含一个或几个药对。深入研究药对的配伍机制对揭示中药复方的配伍规律具有重要意义。当归-川芎是治疗偏头痛的传统活血化瘀药对,然而以往对药对当归-川芎的研究没有把化学成分分析、药代动力学特点和药效学相结合,难以揭示其配伍规律。我们的研究结果表明:1)水回流提取时,阿魏酸不稳定,且对藁本内酯提取率低;乙醇是同时提取主成分阿魏酸和藁本内酯的合适溶剂;2)以主成分阿魏酸和藁本内酯为指标成分,药对当归-川芎的最佳提取工艺为8倍量60%乙醇回流提取3次,每次1.5 h。3)化学成分分析和指纹图谱显示,药对不同配比醇提时,主指标成分阿魏酸和藁本内酯的含量与理论值相比没有明显差异,当归、川芎成分具有加和性,没有新成分生成。4) 大鼠灌胃给予不同配伍比例的当归-川芎成分,主成分阿魏酸和藁本内酯的血浆药物代谢动力学特点呈现一定的规律变化,阿魏酸代谢符合一室模型,藁本内酯代谢为二室模型。5) 当归-川芎对硝酸甘油偏头痛大鼠有治疗作用,其治疗作用与提高脑内5-HT 含量及降低脑内P物质含量相关,但当归-川芎配伍比例与偏头痛疗效没有显著差异。

英文摘要: Herbal pair is a special and relatively simple form of Chinese medicinal formula. Many Chinese medicinal formula contain one or several Herbal pairs. Thorough research of the compatible rules of herbal pair shows important significance in revealing the compatible regularity of Chinese medicinal formula. Angelica Sinensis - Ligusticum chuanxiong is a traditional herbal pair to activate blood and resolve stasis in the treatmentof migraine. However, the previous study of the herbal pair Angelica Sinensis - Ligusticum chuanxiong didn't combine the chemical composition analysis, the pharmacokinetic characteristics and therapeutic effects together. So, It's hard to reveal the compatible rules. Our study resulted in the following aspects. 1)Ferulic acid was not stable and the extraction rate of ligustilide was lower when the herbal was refluxed in water. Ethanol is the suitable solvent for extracting ferulic acid and ligustilide simultaneously from the herbal pair. 2) The optimized technology was that the herbal was refluxed in 60% ethanol by 8 times volume for 3 times and each time 1.5 h, as the main components ferulic acid and ligustilide were considered. 3) The determined contents of ferulic acid and ligustilide showed no difference significantly compared with the theoretical contents. The components of the herbal pair were the total components of Angelica Sinensis and Ligusticum Chuanxiong and with no new ingredients. 4) The plasma pharmacokinetic characteristics showed certain regularity with ferulic acid metabolism of a one-compartment model and ligustilide metabolism of two compartment model, when rats were administrated orally with the components of the herbal pair with different ratios. 5) The herbal pair Angelica Sinensis and Ligusticum showed therapeutic effects on GNT migraine rats and the therapeutic effects were correlated with the increasing content of cerebral 5-HT and the decreasing content of P substances. However, the compatible ratio and the therapeutic effects on migraine were with no significant difference.
