中文摘要: 我们最近研究发现,NRP1在食管癌组织中显著过表达。初步功能分析显示,敲降NRP1可以显著抑制食管癌细胞的侵袭和运动,同时导致NF-κB(P65)表达降低;敲降P65表达,食管癌细胞侵袭运动能力显著降低;而当敲降NRP1回复P65时,细胞侵袭运动能力得到逆转。通过Co-IP分析发现,NRP1可能与HSP60、GRP75等蛋白存在相互作用。但是,NRP1通过NF-κB增强食管癌细胞侵袭运动的分子机制以及NRP1与其相互作用蛋白如何调控NF-κB通路的机制尚不明确。基于此,本项目拟综合运用细胞和分子生物学技术,在细胞水平、动物模型及食管癌标本进一步深入研究NRP1与候选相互作用蛋白的关系、NRP1如何调控NF-κB、以及它们对食管癌细胞侵袭运动的影响,以期揭示NRP1在促进食管癌细胞侵袭运动中的确切作用和详细机制,为阐明食管癌侵袭转移的分子基础及该病的诊断与治疗提供理论依据和分子靶点。

英文摘要: In our previous study, we found NRP1 was up-regulated in esophageal squamous cell carcinoma (ESCC) tissues. Preliminary functional analysis showed that knockdown of NRP1 suppressed migration and invasion of ESCC cells and down-regulated the expression of NF-κB (P65). We also found several potential NRP1 interacting proteins, such as HSP60 and GRP75. However, the mechanisms that NRP1 overexpression enhanced migration and invasion potential through NF-κB (P65) and NRP1 signal transduction need to be further addressed. Basis on the preliminary findings, we will investigate the functional role of NRP1 in ESCC metastasis. This project can reveal the molecular mechanisms of ESCC metastasis, and provide candidate molecular targets for the therapy of the disease.
