中文摘要: 人类表皮生长因子受体2(HER2)是乳腺癌重要的分子标志之一,HER2阳性乳腺癌具有恶性程度高,侵袭性强,预后差等特征。目前,临床HER2表达检测缺乏特异性、时效性和全面性,靶向检测HER2的分子成像技术为乳腺癌的鉴别诊断、预后评估、治疗方案的选择提供了新方法。本课题优化HER2亲合体多肽设计,一步法便捷制得特异性识别HER2分子探针18FAl-NOTA-MAL-Cys-M0/59ZHER。采用体外分析和体内microPET显像、生物分布等筛选出合适的分子探针,考察该新型HER2分子影像探针用于乳腺癌HER2的在体定位、定量及可视化的可行性。荷瘤裸鼠行抗HER2药物治疗,应用分子影像技术结合病理学等综合分析该探针用于早期判定肿瘤靶向治疗疗效的前景。本项目若获成功,将为乳腺癌临床诊疗提供新思路,也为开发新型的肿瘤诊治方法奠定了基础。

英文摘要: Human epithelial growth fator receptor-2(HER2) is an important marker of breast tumor. HER2 positive breast cancer is associated with increased tumor aggressiveness, resistance to therapies and poor outcome. Currently, determination of HER2 status is lack of specificity, chronergy and comprehensiveness. Molecular imaging targeting HER2 provides a new method for differentiated diagnosis, prognosis and choosing therapeutic scheme in oncology. In this subject, after optimum design an affibody, a specific HER2 probe 18FAl-NOTA-MAL-Cys-M0/59ZHER was easily obtained from one-step reaction. In vitro cells experiments , microPET imaging, biodistribution study were performed for screening a optimal probe. The feasibility of in vivo location, quantitative measurement and visualization of HER2 in breast tumor with the probe was also evaluated . Tumor bearing mice were treatment with anti-HER2 drugs. The prospects of early assessment the effects of target-therapy on tumors by the probe were analyzed through molecular imaging combing with pathology. If successful, the subject will provide a new technology for diagnose and therapy of breast tumor.
