中文摘要: 基于抗凋亡蛋白为靶点设计抗肿瘤药物是目前一个热门研究领域。另外,人血清白蛋白由于自身的特性,作为抗肿瘤药物的载药体系得到广泛地研究。因此,本项目融合生物化学、无机化学、结构生物学、分子药剂学和同步辐射技术等开展以下研究:(1)无机药物与抗凋亡蛋白(Bcl-2)相互作用的生物学基础(2)白蛋白结合单一/多种无机药物的生物学基础(3)白蛋白-无机药物复合物的抗肿瘤行为。本项目将进一步理解无机药物促进肿瘤细胞凋亡的机制,阐明白蛋白递送单一/多种无机药物的结合特征,揭示无机药物-白蛋白复合物的抗肿瘤效应及其机制;无机药物-蛋白之间的定量构效关系。旨在为开发新型抗肿瘤无机药物,白蛋白作为抗肿瘤无机药物的多功能载体提供重要生化数据和理论基础。

英文摘要: Currently,anti-apoptosis protein-based design of anticancer drugs is hot research area. Furthermore, human serum albumin(HSA) drug delivery systems have been extensively studied owing to its properties. Therefore, the project will carry out the following research by combining biochemistry、inorganic chemistry、structural biology、molecular pharmaceutics with synchrotron radiation source.(1) the biochemical basis of interactions between anti-apoptosis protein (Bcl-2) (2)the basic of biology of single/multi drugs binding to HSA (3) The anti-cancer behavior of HSA-inorganic drugs complexes. The project will promote us to understand the mechanism on how inorganic drugs induce apoptosis of cancer cells, reveal the binding properties of HSA delivering single/multi drugs, the anticancer of efficacy and mechanism of HSA-inorganic drugs complexes; quantitative structure-activity relationship between inorganic drugs and protein. Our object is to develop new type inorganic antitumor drugs, provide important biochemical data and theoretical foundation for HSA-based multifunctional carriers for inorganic anti-tumor drugs.
