中文摘要: 卵巢过度刺激综合征(OHSS)是辅助生殖技术中较严重的一种并发症,其发病机制目前尚不清楚。抗苗勒氏管激素(AMH)能够抑制始基卵泡的活化和发育,降低卵泡对FSH的敏感性、抑制颗粒细胞生长,抑制颗粒细胞中芳香化酶活性。AMH信号传导通路是卵泡发育的抑制机制之一。前期研究中,我们发现OHSS患者的AMH基因存在SNP突变。因此,我们推测AMH信号传导通路在OHSS发生发展过程中发挥了重要的作用。为证明这种假说,本实验应用分子生物学、基因测序、组织化学等技术,探讨AMH信号通路各个环节在OHSS患者颗粒细胞中的表达规律及其在OHSS发生发展过程中的作用和机制。本研究将从卵泡发育抑制机制异常的角度,进一步解释OHSS发生发展的分子机制,为寻求有效的OHSS防治方法提供新的理论依据和新的靶点。

英文摘要: Ovarian hyperstimulation syndrome (OHSS)is a severe complication observed in some patients undergoing controlled ovarian hyperstimulation (COH) during assistened reproductive medicine,and its mechanism is unclear. Anti-Mullerian hormone (AMH) may play an inhibitory role in the development of follicles, including:1)inhibition of follicular activation and growth; 2)inhibition of FSH stimulated growth; 3)inhibition of GC growth; 4)and inhibition of aromatase. During our previous study, we found that SNP mutation exist in the AMH DNA. Thus, we presume that AMH signal pathway may play an important role during the nosogenesis of OHSS. To prove this hypothesis, we use molecular biologic technology, DNA sequencing and histochemical technology, observing the expression of AMH signal pathway in the GC of OHSS patient and the role of AMH signal pathway during the nosogenesis of OHSS. By this study, we can understand the nosogenesis of OHSS more clear, and this study may provide us a new theory and target to prevent and cure OHSS.
