中文摘要: 生物碱是一类便宜易得的天然产物,与生俱来的酸、碱官能团并存形式注定它们在不对称有机催化中占据着非常重要的地位。本项目拟设计合成一类在生物碱碳-9位引入Br?nsted酸官能团并在碳-9位以非醚键联接的系列新型C2对称生物碱二聚物,建立催化剂库。这些生物碱二聚物将作为Lewis碱催化几类不对称烯丙基烷基化反应;作为Br?nsted碱催化以β,γ-Butenolides为底物的几类不对称反应。研究中将充分利用该类型生物碱二聚物的Linker、氢键给体及生物碱单体可调性强的特点,探索出各类反应取得高立体选择性的催化剂及反应条件。通过研究催化剂在不同反应中的手性控制机理,明确在生物碱二聚物中引入Br?nsted酸对于其分别作为Lewis碱与Br?nsted碱催化剂的影响,从中找到一些规律。本课题的开展将丰富C2对称生物碱二聚物的类型,为其在有机小分子不对称催化中的应用起到推动作用。

英文摘要: Cinchona alkaloids, which contain both acid and base founctional groups, are natural asymmetric catalyst with widespread applications in organic synthesis. The project is designed to introduce Br?nsted acid moiety, which has been demonstrated as remarkable versatile catalytic groups in organic reactions, into Cinchona alkaloids at C-9 position to synthesize novel C2-symmetric (bis)cinchona alkaloid asymmetric catalyst. The project will also evaluate the applications of C2-symmetric (bis)cinchona alkaloid asymmetric catalyst as Lewis base or Br?nsted base to varies substrates, such as, Morita-Baylis-Hillman carbonates, β,γ-Butenolides. Apporiate modification of the linker, hydrogen bonding group as well as the alkaloid base will help to explore suitable reaction with excellent enantioselectivities. We will also invistage the machanism of the reaction pathway to determine the invovlement of the catalyst as Lewis base or Br?nsted base. The project will enrich the C2-symmetric (bis)cinchona alkaloid asymmetric catalyst pool; it will also contribute to explore new asymmetric reactions.
