中文摘要: 常规近场光学显微镜的成像受限于所用的针尖,即有孔型光纤尖端的弱信号和无孔型针尖散射光偏振信息的不完整。基于表面等离激元的新型近场光学显微镜则可以突破这些局限,实现高强度局域照明、出射光的保偏、高空间分辨率等功能。金属纳米线中激发的表面等离激元具有局域电磁场增强、传播不受衍射极限的限制以及偏振状态可控等特点,在纳米光源、生物细胞检测和分子手性识别方面具有潜在的应用价值。本项目的实施将主要围绕仪器整体规划、部件设计与组装、样品表征与技术指标验证这三个方面,同时展开对相关基本科学问题的研究。我们将首先定位本项目的基本技术指标,从而进行部件设计和采购。集中解决两大关键技术问题,即金属纳米线探针的制作和系统稳定性及弱光学信号的探测。最后我们将采用标准样品对仪器进行技术指标验证,并尝试扩展到复杂样品,如生物细胞样品和手性单分子样品,研究细胞内部分子结构,以及利用纳米光源的手性偏振对分子进行手性识别。

英文摘要: Conventional near-field optical microscopic imaging is limited by the optical probes, that is, the weak signals in aperture type fiber tips and polarization unintegrity of the scattered light by apertureless tips. Novel near-field optical microscope based on surface plasmons supported by metal nanowires can overcome such limitations, facilitating functionalities such as nanoscale illumination with high intensity, polarization reservation and high spatial resolution. Surface-plasmons supported by metal nanowires have unique characteristics including local electromagnetic field enhancement, propagation beyond the diffraction limit and controllable polarization status, implying important applications in nanoscale light source, cell detection/therapy and chirality identification in biology. This project will cover equipment master plan, accessories design and assembly, sample characterization and technical specifications assessment. The meanwhile, related scientific problems will also be investigated. We will first establish the technical specifications for the performance of the equipment, and then proceed with corresponding accessories design and purchase.
