中文摘要: 结核病是危害人类健康的重大疾病,其广泛耐药性是目前治疗的主要屏障。研究显示结核分枝杆菌的适应性免疫应答是结核感染具有持久性和易耐药的原因之一。本项目探讨结核分枝杆菌适应性免疫应答的发生机制,观察亚日饮汤对结核分枝杆菌感染细胞免疫应答的调节作用。用结核分枝杆菌感染C57BL/6J及其巨噬细胞,观察药物干预后巨噬细胞功能、T细胞介导的细胞免疫包括树突细胞亚群、T细胞亚群、细胞因子(IFN-γ、IL-17、IL-23、IL-12和TNF-α等)、PD-1/PD-L免疫调节信号途径的改变;制备抗 PDL1 抗体,观察干扰PD-1/PD-L信号系统联合亚日饮汤治疗对T细胞免疫的影响,检测小鼠脾脏淋巴细胞IFN-γ分泌水平,脾脏CD8+T细胞PD-1表达情况。为阐明结核分枝杆菌适应性免疫应答的发生机制与防治药物的研究提供实验数据。

英文摘要: Tuberculosis is a major disease hazard to human health, which is the main barrier to extensively drug-resistant. Research shows that adaptive immune response to Mycobacterium tuberculosis is one of the reasons to persistent and drug-resistant. This project explores the mechanisms of adaptive immune response to Mycobacterium tuberculosis and observes regulation of Yari Yin Tang on cellular immune response infected with Mycobacterium tuberculosis. To Infect macrophages and C57BL/6J mice with Mycobacterium tuberculosis and drug-resistant mycobacterium tuberculosis to isoniazid and rifampicin.Observe macrophage function, cellular immune response subseted by T cell, including dendritic cells and T cells, cytokines (IFN-γ , IL-17, IL-23, IL-12 and TNF-α, etc.) and PD-1/PD-L pathway after drug intervention. To product antibody to anti-PD-L 1. Observe the effect on T cell immunity with interference PD-1/PD-L system and combined with Yari Yin Tang. Detect IFN- levels in splenic lymphocyte and the expression of PD-1 in splenic CD8+ T cell. Provide experimental data for elucidating the mechanism of the adaptive immune response to Mycobacterium tuberculosis and prevention and research of drug research .
