中文摘要: 花青素具有抗氧化、抗炎及抗衰老等许多生物活性,其抗肿瘤作用研究方面近年来备受关注。课题组前期研究表明,蓝莓花青素能抑制KB细胞的增殖,阻滞细胞周期于G2/M期;用浓度为50、100、200μg/ml蓝莓花青素处理KB细胞,浓度50μg/ml时,p53不表达,当浓度达到100μg/ml时,p53开始表达。p53的活化会影响细胞周期阻滞、凋亡、代谢、分化及自噬等多种生理活性,我们选取了自噬和凋亡作为本研究的切入点,在前期工作基础上,采用动物模型和体外实验相结合的方法,从细胞、动物模型及分子水平上,观察蓝莓花青素对口腔癌细胞及裸鼠动物模型自噬和凋亡的影响,并探讨蓝莓花青素抗口腔癌作用的相关分子机制。该项目的研究将对内蒙古自治区特色资源--野生蓝莓的开发利用具有重要的意义,并为临床应用蓝莓花青素防治肿瘤及其药效学评价提供理论依据和新的治疗模式。

英文摘要: The bioactivity of anthocyanins includes antioxidant, anti-inflammatory, anti-aging and so on. Our prior studies revealed blueberry anthocyanins can significantly inhibit cell proliferation of oral cancer and block the cell cycle in the G2/M period. KB cells were treated by 50,100,200μg/ml blueberry anthocyanins. The results showed that p53 was not expressed at concentration of 50μg/ml, but expressed at concentration of 100μg/ml.Activation of p53 affected much physiological activity such as cell cycle arrest,apoptosis, metabolism, differentiation and autophagy.In this study,oral cancer cells and animal model were used to explore the anti-cancer effect and mechanisms of antophagy and apoptosis induced by blueberry anthocyanins in oral cancer.It will provide certain theoretical and experimental basis for development and utilization of blueberry which is characteristic resources of Inner Mongolia and clinical application blueberry anthocyanins to prevent and treat oral cancer.
