中文摘要: Protrudin是近年新发现与神经轴突生长密切相关的一个蛋白,它通过调节囊泡运输诱导神经轴突的形成。protrudin作为酸性亚细胞器-内体(endosome,pH ~5)的膜蛋白,在调节胞内运输过程中势必与许多蛋白协同发挥作用,但受限于常规的蛋白相互研究技术的局限性,很难在酸性条件下研究蛋白相互作用。针对这一问题,我们将"基因密码子扩展技术"应用于蛋白相互作用研究领域。本项目利用该项技术,将具有光交联活性的非天然氨基酸定点引入到protrudin蛋白中,通过光照交联,捕捉protrudin新的互作蛋白,经质谱鉴定其中一个互作蛋白是微管相关蛋白4(MAP4)。进而通过基因过表达、基因沉默等技术,在分子、细胞、动物(斑马鱼)水平,研究protrudin和MAP4的相互作用及功能,揭示神经轴突生长的分子机制及网络调控,为神经轴突变性的治疗提供新的靶点或理论支持。

英文摘要: Protrudin is a new protein associated with neurite outgrowth, which regulate vesicle trafficking to induce neurite formation. There must be many other proteins involved in protrudin regulated intracellular material transport, as protrudin is a membrane protein of acidic endosome (pH ~5). However, it is very difficult to study protein-protein interaction in acidic condition under the traditional technology. To solve the problem, "genetic code expansion technology" was used to research protein-protein interaction. In this study, the unnatural amino acid with photo crosslinking activity was site-specifically inserted into protrudin. Through photo crosslinking and mass spectrometry analysis, microtubule associated protein 4 (MAP4) was found as a new interaction protein of protrudin. Then, we plan to study role of the interaction between protrudin and MAP4 in neurite outgrowth at molecular, cellular and whole animal (zebrafish) levels. We intend to reveal the molecular mechanisms regulation network of axonal growth providing new target or theoretical support for the treatment of axonal degeneration.
