中文摘要: 大豆-根瘤菌共生固氮是农业生态系统中重要的氮素来源,微量元素铁既是固氮关键酶的组分,又是大豆根瘤中豆血红蛋白的组成元素,对根瘤的生长发育及固氮尤为重要,然而土壤缺铁是大豆生产中面临的养分难题之一,如何提高共生系统中大豆对铁的利用效率是植物营养学关注的热点。本项目利用已报道的大豆基因组和表达谱数据库,确定参与二价铁吸收运转的NRAMP基因家族共有13个成员。通过组织表达特异性分析,筛选出根瘤特异表达的GmNRAMP2基因,将进一步通过组织定位和功能分析,探讨该基因在大豆根瘤铁转运中可能的功能。本研究不仅对探讨NRAMP基因在维持大豆根瘤铁稳态中的功能研究具有理论意义,也为豆科作物的分子改良提供重要的理论依据。

英文摘要: Soybean-Rhizobium symbiosis is the important nitrogen(N) source in agroecosystems,in particular,iron is very important for nodule development and N fixation, because it is not only an important component of nitrogenase, but also the element of leghenmoglobin in soybean nodules. However, iron deficiency is one of the nutrient problems in practice. How to improve iron utilization efficiency in symbiosis system is a focus of plant nutrition research. In this proposal, we had been identified and isolated 13 NRAMP genes in the soybean genome based on reported soybean genomic DNA database and expression profile.Through tissue-expression pattern analysis, nodule-specific gene GmNRAMP2 had been chosen as candidate gene,tissue localization and functional analysis will proceed to investigate the possible role of GmNRAMP2 in iron transport of soybean nodule. This research will not only provide theoretical significance on the functions of NRAMP gene in nodule iron nutrition, but also give theoretical basis for molecular improvement in legume crops.
