中文摘要: 生长因子活性及缓释体系是决定骨诱导效能的关键因素。申请人博士期间分别探讨了骨形态发生蛋白BMP2/7异源二聚体对成骨细胞和破骨细胞的作用特点以及体内应用研究。与BMP2、BMP7同源二聚体相比,BMP2/7具有起效时间短、有效剂量低等特点,能够高效诱导骨再生。此外,申请人留学期间参与磷酸钙缓释系统的研究,证实该系统能与多种生长因子共沉积,实现其体内缓慢释放,从而最大限度优化其生物效能,并且该系统适用于多种生物材料载体。然而,BMP2/7在磷酸钙缓释系统中可发挥的骨诱导效能,及其对成骨-破骨偶联作用的影响尚不明确。本课题将BMP2/7与缓释技术相结合,通过成骨细胞-破骨细胞共培养体系来探讨BMP2/7缓释系统对成骨-破骨这一偶联体系的作用及机制;同时明确缓释系统是否能促进BMP2/7的骨诱导效能。本研究将为BMP2/7缓释系统在体内高效、快速诱导成骨,促进骨改建提供一定的理论依据和研究基础。

英文摘要: The bioactivity of growth factors and controlled release system are crucial for osteoinductive efficiency. We investigated the functional characteristics of bone morphogenetic protein (BMP) 2/7 heterodimer on osteoblastogenesis and osteoclastogenesis respectively. In comparison to BMP2 or BMP7 homodimers, BMP2/7 heterodimer showed a more rapid effect with significantly lower effective concentrations, thus could highly efficiently induce bone regeneration. We also previously corroborated that biomimetic calcium phosphate coatings could coprecipitate with many growth factors and realize their in-vivo controlled release. However, its effect in controlled release system on the coupling between osteoblast and osteoclast remains unclear. In this proposal, we will adopt a co-culture system to investigate the influence of BMP2/7 heterodimer on the coupling of osteoblast and osteoclast. Meanwhile, we will use biomimetic calcium phosphate coating to coprecipitate and controlled release BMP2/7 heterodimer so as to further increase its osteoinductive efficiency. We will establish a new research line in using BMP2/7-coprecipitated control release system for highly efficient bone induction.
