中文摘要: 植物分泌结构的发生发育和衰老过程与次生代谢产物的产生和积累有着非常密切的关系。以往有关植物分泌结构的研究主要集中在其发生发育、衰老,及分泌物的产生与特定细胞器的关系等方面,而忽略了分泌结构发生发育过程中分泌物的积累及与分泌细胞降解的关系研究。为此,本项目拟应用植物解剖学、细胞化学、显微免疫荧光标记和分子生物学技术,以芸香科植物白鲜分泌囊、头状腺毛和囊状腺毛为研究对象,系统研究分泌结构的发生、发育和衰老过程及挥发油类物质在分泌细胞中产生、转运和积累规律;深入研究挥发油积累过程中分泌细胞PCD的显微及超微结构变化、细胞核和基因组DNA的降解特点、降解酶的活性变化;从显微、超微结构及分子水平阐明白鲜分泌结构挥发油的产生和积累与其发生发育及PCD过程的关系。研究结果不仅可为植物分泌结构次生代谢产物的产生和积累方式提供新的证据,也为植物次生代谢产物调控的进一步研究提供科学依据。

英文摘要: The developmental and degeneration process of plant secretory structures have shown close relationship with secretion synthesis and accumulation in present studies. In current literature, extensive microscopic examination on the morphological, developmental and ultrastructural details of plant secretory structures has been referred to, however a detailed study on the relationship between the secondary metabolite synthesis and the degradation process of plant secretory structures is still lacking, which leads to different views expressed among early investigators concerning the mechanism of secretion. In the present work, histochemistry, fluorescence immunoassay, DNA ladder, cytological chemistry, DNA SDS-PAGE and ultraviolet spectrophotometry are applied to determine the mechanism of development and lipid synthesis, nuclease and caspase-3 like activity, degeneration of nucleus, nucleic acid fragmentation as well as cytological characters of secretory structures in Dictamnus dasycarpus. The aims are to clarify the mechanism of lipid synthesis and accumulation in the secretory structures of Dictamnus dasycarpus from cytological points of role, to reveal the relationship between degradation product of PCD and lipid synthesis in the secretory structures of Dictamnus dasycarpus, and to provide new views and evidence
