中文摘要: Cbx4是多梳抑制复合物PRC1家族的一个重要成员。最近,我们实验室报道Cbx4能够通过其SUMO化E3连接酶的活性使低氧诱导因子1a(HIF1a)发生SUMO化修饰从而增强肝癌的新生血管形成。在此基础上,我们新近发现Cbx4是肝癌病人预后的一个独立因素。Cbx4高表达的病人经过动脉栓塞治疗(TACE)后能显著延长其生存期。进一步地,我们发现Cbx4能够增强肝癌细胞对于化疗药物的敏感性。基于这些前期工作,本课题拟深入研究Cbx4增强肝癌细胞对于化疗药物敏感性及其具体的分子机制,通过临床与基础研究相结合,以期为临床肝癌病人的治疗提供一定的理论依据和线索。

英文摘要: Cbx4 is a family member of polycomb repressive complexes 1(PRC1). Recently, our group reported that Cbx4 can govern HIF1a to potentiate angiogenesis of hepatocellular carcinoma by its SUMO E3 ligase activity. On this basis, we discovered Cbx4 is an independent prognostic factor of hepatocellular carcinoma.Transarterial chemoembolization (TACE) treatment can prolong survival of Cbx4 high-expression patients. Furthermore, we found that Cbx4 could enhance the sensitivity of the liver cancer cells to chemotherapy drugs. Based on these findings, we intend to explore the molecular mechanism of how Cbx4 enhance the sensitivity of hepatocellular carcinoma to chemotherapy drugs.Through the combination of clinical research and basic research, we may provide theoretical basis and clues to select treatment methods of liver cancer patients.
