中文摘要: 中枢敏化PKC/VGCC/ Ca2+/Glu/NR2B离子通道是内脏痛敏发生、发展过程的关键因素。基于课题组前期研究,本项目拟以内脏痛敏IBS大鼠为研究载体,分离并培养其脊髓背根神经元和结肠肥大细胞,应用全细胞膜片钳、激光共聚焦显微技术、活细胞工作站等当今先进技术①探讨内脏痛敏IBS大鼠PKC/VGCC/Ca2+/Glu/NR2B离子通道在内脏痛敏启动效应中的作用机制。②通过比较温和灸与鞘内分别注射选择性N型VGCC拮抗剂齐考诺肽、NR2B拮抗剂CP101606对内脏痛敏IBS大鼠的治疗,探讨温和灸对中枢敏化PKC/VGCC/Ca2+/Glu/ NR2B离子通道的调控作用,从中枢探讨IBS内脏痛敏的神经信息传导机制及艾灸对中枢-肠神经信息交流的影响,揭示艾灸治疗IBS内脏痛敏的科学基础。以冀深入揭示艾灸镇痛的科学基础,为应用艾灸治疗痛证的推广,提供实验数据和理论依据。

英文摘要: The central sensitive ion passage of PKC/VGCC/ Ca2+/Glu/NR2B is the key way that related to the onset and development of visceral hyperalgia. In order to discussing the central neural information transmission of visceral hyperalgia, to discussing the warm-moxibustion effect on the central-colon neural information transmission, discovering the scientific basis of analgesia effect of warm-moxibustion on the visceral hyperalgia, to revealing the scientific basis of analgesia effect of moxibustion, providing the experimental data and theory for the application and carring forward the moxibustion therapy, based on our previousely expremental outcome, established the visceral hyperalgia pain rats' model, seperated and cultured the dorsal horn neurons cell and the mast cell of them, the research aims at(1) exploring the PKC/VGCC/ Ca2+/Glu/NR2B ion passage's priming effect on visceral hyperalgia rats.(2)exploring the regulative effect of warm-moxibustion on PKC/VGCC/ Ca2+/Glu/NR2B ion passage by comparing the therapeutic effect of warm-moxibustion with the antagonist of N-VGCC(ziconotide)and/or the antagonist of NR2B(CP101606)on visceral hyperalgia rats by the modern experimental assay such as patch clamp recording technique,laser scanning confocal microscope and living cell working system.
